Lou was member of Marge Simpson's family. When Ned Flanders asks Homer to babysit Rod and Todd, he tells Ned that Marge is identifying her Uncle Lou's body at the city morgue. At the morgue, the body is revealed to be Hans Moleman (who was still alive). However, when Hans Moleman talks, the doctor tells Marge that was just gas escaping.

Don't confuse Uncle Lou with Lou Simpson, despite having the same name, they are different people.
Lou Simpson is Homer's great-granduncle, that is, he is a member of the Simpson family.
Uncle Lou is related to Marge, being part of the Bouvier family or the Gurney family.
